Abstract

Objective: Fetuin-A is a glycoprotein synthesized mainly in the liver and adipose tissue and then released into the bloodstream. Fetuin-A plays important roles in tumor progression, calcification, diabetes, obesity, kidney diseases and coronary artery disease through different mechanisms. Fetuin-A is taken into tumor cells by endocytosis, and while inside the cells, it increases the secretion of exosomes. Lately a number of studies have shown that a subset of tumor cells such as prostate, pancreatic and glioblastoma multiform synthesize ectopic fetuin-A, which promotes their progression. Cisplatin is an anti-cancer chemotherapy drug. This medication is classified as an alkylating agent. It is used to treat a number of cancers. These include breast cancer, testicular cancer, bladder cancer, mesothelioma, cervical cancer, ovarian cancer, lung cancer, head and neck cancer, esophageal cancer, brain tumors and neuroblastoma. The aim of this study was to investigate the effect of cisplatin on the fetuin A level in the SH-SY5Y cell line.

Method: In this study, the SH-SY5Y human neuroblastoma cell line was used. Two cell groups were prepared to investigate the effect of cisplatin on the fetuin A level. The control group did not undergo any treatment. Cells in the cisplatin group were treated with 10 μM cisplatin for 24 hours. Cells were frozen and re-thawed 3 times to burst. It was then centrifuged and the supernatants were collected. After that ELISA (enzyme-linked immunosorbent assay) method was applied to determine the level of fetuin A. Statistical evaluation of the data was performed by independent t-test, Statistical significance was defined at p<0.05.

Results: There was a significant increase in fetuin A level in the cisplatin group compared to the control. (p <0.05).

Conclusions: Cisplatin was found to increase the level of fetuin A in the SH-SY5Y cell line.
